Each capsule contains Acemetacin 90 mg.
Acemetacin belongs to a group of medicines called non-steroidal anti-inflammatory drugs (NSAIDs). It is easily absorbed upon oral administration. It is metabolized to indomethacin by the liver and works by blocking various chemicals in the body known as prostaglandins.
Pharmacology: Acemetacin is a non-steroidal anti-inflammatory drug (NSAIDs) with anti-inflammatory, analgesic and antipyretic properties. It blocks the action of COX and reduces the production of prostaglandins, which causes pain and swelling.
Pharmacokinetics: Acemetacin is easily absorbed upon oral administration, protein binding is 81.5, 93.7% and is metabolized to Indomethacin in the liver, which becomes the active metabolite. The main excretion is eliminated by the kidney, partial excretions are by the bile and feces.
After taken 8 consecutive days of ACEO Capsules Retard, the time to reaches the maximum blood concentration is average of 4.5 hours. And, the average half-life of Acemetacin is about 4.8 hours. The average half-life of Indomethacin is about 8.2 hours.
Rheumatoid Arthritis, degenerative arthritis (osteoarthritis), ankylosing spondylitis, gout.
Unless advised specifically by a medical doctor, the adult dosage is one capsule daily. For acute cases, and depending on the patient's condition, take 1 capsule twice daily. For acute gout, take 180 milligrams (one capsule twice daily) everyday, until symptoms disappear. Swallow whole with water, after meals (do not bite). Adjust dosage based on severity of condition. For acute cases and acute gout required high dosage, should not take this drug more than 5 consecutive days.
Do not take if the patient is allergic to Acemetacin or to any preparation with Indomethacin.
Blood coagulation disorder.
Gastrointestinal ulcers.
Pregnant and lactating women.
Children under 16 years old.
Do not use in children under 16 years old, pregnancy, lactating women, peptic ulcer or hemorrhage.
Avoid using in patient with vascular disease risk in order to reduce risk of cardiovascular, cerebrovascular and peripheral thrombotic events.
Avoid long term use due to increased risk of peptic hemorrhage, hepatic dysfunction, renal dysfunction unless advised specifically by a medical doctor.
In case of vertigo, do not drive or operate dangerous machinery. Drinking alcohol may increase dizziness.
Patients with history of peptic or duodenal ulcers must not use this drug. In case of necessity, take only under the strict supervision of a medical doctor.
Use with caution if patients have liver or kidney disease under the strict supervision of a medical doctor.
Do not administer to patients with blood abnormalities. In case of necessity, take only under the strict supervision of a medical doctor.
This medicine should be administered with caution if patient has high blood pressure, heart disease (heart function disorder) or had just undergone a major surgery. Caution should also be exercised when decreasing dosage or shortening intake period. Take only under the strict supervision of a medical doctor.
Caution should be taken if a patient is allergic to any NSAIDs; have experienced in asthma, skin rashes, or any allergic reactions, chronic obstructive airway disease, hay fever and nasal polyps.
It is not known how this drug would affect blood coagulation and blood sugar, when taken in combination with anticoagulants and blood-sugar lowering drugs.
Patients receiving long-term treatment should be periodically screened for renal and hepatic function and blood counts. If a surgical procedure is necessary, consult with the doctor on whether to continue intake.
If diuretics are being used simultaneously, monitor content of potassium or lithium salt in blood.
Taking this medication may aggravate psychiatric disorders, epilepsy, and Parkinsonism.
Use in Children: Children under 16 years old should avoid taking this drug.
Use in the Elderly: If necessary, this medicine may be administered to the elder with caution and only under the strict supervision of a medical doctor.
Pregnant and lactating women: Should not be taken before and during pregnancy, unless absolutely necessary. Using this medication during the last three months of pregnancy is dangerous to both mother and child. It may cause complication of liver, kidney and intestines. A small quantity of Acemetacin and its metabolites are found in the milk and lactating women taking this drug. It is therefore not to be taken before giving birth and during lactation.
Gastrointestinal: Nausea vomiting, abdominal pain, diarrhea and gastrointestinal perforation and hemorrhage. May have discomfort, ingestion, bloating, abdominal cramps, loss of appetite, peptic ulcer and bleeding. Occasionally, may experience blood in sputum, mouth sores, ulcers in esophageal lining, intestinal bleeding, enteritis, constipation. If the patient suffers from acute abdominal pain, black stool, stop the medication and see the doctor immediately.
Central nervous system and sensory organs: May experience headache, agitation, anxiety, fatigue, confusion, depression, tinnitus, dizziness, loss of consciousness, complete comatose, myasthenia, profuse perspiration, nervous breakdown, paranoia, borderline neurosis, cerebral palsy, shortage of memory, unusual taste sensory and abnormal vision (blurred vision, double vision, sees black spot and halo), hearing impairment, memory loss, convulsions, insomnia, nightmare, trembling, etc. Long term use may cause retinal pigmentation, corneal erosion and blurred vision.
Liver and pancreas: Lesion on the liver, pancreatitis, increased blood sugar, diabetes.
Kidney and urinary system: Kidney damage, albuminuria, blood in the urine. Stop taking the medicine as soon as symptoms occur or worsen, and consult a doctor immediately.
Heart circulatory system: High blood pressure, discomfort and palpitation.
Allergy and skin reaction: Skin rash, itchiness, urticaria, mouth sores, alopecia, rare blisters, eczema, dermatitis, bleeding, Stevens-Johnson Syndrome, Lyell's Syndrome. Swelling, dropping eyelids, swelling of tongue, throat inflammation, breathing difficulty, palpitation, low blood pressure, shock, etc. See the doctor immediately if symptoms appear. Anaphylaxis or pneumonia may happen.
Blood: Blood abnormality such as anaemia, high leucocytes count; low blood platelets count. Initial symptoms would be fever, headache, mouth sores, colds, fatigue, nosebleed, skin bleeding.
Others: Blood in urine, increased potassium in blood. Hypertensive patient may have kidney dysfunction, and edema. NSAIDs have been reported to cause nephrotoxicity. In case of the following reactions: redness, swelling, hot flashes, pain, fever, etc., stop taking the medication and see a doctor immediately.
Blood concentration of Acemetacin may be affected if it is taken with cardiac stimulants, convulsion medicine (phenytoin), and lithium.
Drug efficacy of hypertensive drugs may decrease with this medication. It can also rapidly increase the elimination process of the diuretic furosemide from the body.
This drug may decrease the antihypertensive effect of ACE inhibitors and cause renal dysfunction.
Potassium level in blood may increase if diuretic is used with this drug.
This drug and other NSAIDs have gastrointestinal side effects.
This drug is contraindicated with chemotherapy drug methotrexate.
Gout medicine, probenecid or sulfinpyrazone can inhibit the excretion of this drug.
Taking this drug with blood coagulants increase danger of hemorrhage. It is necessary to monitor blood coagulation when combining these drugs.
This drug taken with cyclosporine can increase kidney toxicity.
Taking this drug with aspirin may decrease its blood concentration.
Caution should be exercised when taking this drug with central neural agents and alcohol.
This drug should not be used together with diuretic triamterene as indomethacin may cause acute renal dysfunction.
This drug should not be taken with diflunisal, as it will increase the concentration of indomethacin and cause fatal stomach hemorrhaging.
Elimination of penicillin is delayed when used with this drug.
