Symptomatic treatment of chronic constipation in adults whom laxatives fail to provide adequate relief.

Adult 2 mg once daily. Elderly >65 yr Initially 1 mg once daily, may be increased to 2 mg once daily if needed. Severe renal impairment (GFR <30 mL/min/1.73 m²) 1 mg once daily.

May be taken with or without food.

Hypersensitivity. Patients w/ stomach pain, nausea or vomiting. Intestinal perforation or obstruction due to structural or functional disorder of gut wall, obstructive ileus, severe inflammatory intestinal tract conditions (eg, Crohn's disease, ulcerative colitis & toxic megacolon/megarectum). Renal impairment requiring dialysis.

Discontinue therapy if patient develops severe, persistent, &/or worsening abdominal symptoms, bloody diarrhea or rectal bleeding. Patients w/ severe & clinically unstable concomitant disease eg, CV or lung disease, neurological or psychiatric disorders, cancer or AIDS & other endocrine disorders. Patients w/ history of arrhythmias or ischaemic CV disease. May reduce efficacy of OCs in case of severe diarrhoea; use additional contraceptive method. Not to be taken by patients w/ rare hereditary problems of galactose intolerance, total lactase deficiency or glucose-galactose malabsorption. Minor influence on ability to drive & use machines. Reconsider treatment in patients w/ hepatic impairment if once daily dosing is not effective after 4 wk. Women of childbearing potential must use effective contraception during treatment. Not recommended during pregnancy & in women of childbearing potential not using contraception. Lactation. Not recommended in childn & adolescents <18 yr.

Headache; nausea, diarrhoea, abdominal pain. Decreased appetite; dizziness; vomiting, dyspepsia, flatulence, abnormal GI sounds; fatigue.

Increased plasma conc of erythromycin. Increased systemic exposure w/ ketoconazole. Concomitant use w/ potent P-gp inhibitors eg, verapamil, cyclosporine A & quinidine.
