Metformin HCl
Indications/Uses
Monotherapy or in combination w/ sulfonylurea in treatment of NIDDM patients who are no longer responding to sulfonylurea.
Dosage/Direction for Use
Adult Monotherapy & combination w/ other oral antidiabetic agents Initially 500 or 850 mg 2-3 times daily. May be slowly increased by an increment of 1 tab depending on blood glucose level after 10-15 days. Max: 3 g daily in 3 divided doses. Combination w/ insulin Initially 500 or 850 mg 2-3 times daily, while insulin dosage is adjusted on the basis of blood glucose measurements. Childn ≥10 yr & adolescents Monotherapy & combination w/ insulin 500 mg daily. Max: 2 g daily in 2-3 divided doses.
Administration
Should be taken with food: Take w/ or after meals.
Contraindications
IDDM, ketoacidosis, severe infection, stress, trauma & other severe conditions w/ uncontrolled hyperglycemia. Should not be given in patients w/ severe renal or hepatic impairment. Heart failure, recent MI, dehydration, acute or chronic alcoholism & other conditions predisposing to lactic acidosis.
Special Precautions
Lactic acidosis. Should not be given to pregnant or lactating women.
Adverse Reactions
Impaired vit B12 absorption; taste disturbance; nausea, anorexia, diarrhea; pruritus, erythema, urticaria.
Drug Interactions
Risk of lactic acidosis w/ cimetidine, diuretics, alcohol & alcohol-containing medications.
