Mitomycin

Indications/Uses

Disseminated adenocarcinoma of stomach or pancreas w/ other chemotherapeutic agent therapy & as palliative treatment when other modalities fail.

Dosage/Direction for Use

Hematological recovery from previous chemotherapy 20 mg/m² IV as single dose at 6-8 wk interval.

Contraindications

Hypersensitivity or history of idiosyncratic reaction. Patients w/ thrombocytopenia, coagulation disorder or increased bleeding tendency due to other causes.

Special Precautions

Monitor platelet & WBC count, differential & Hb repeatedly during therapy & at least 8 wk. Withhold further therapy if platelet count & WBC are below 100,000/mm³ & 4,000/mm³, respectively. Pregnancy.

Adverse Reactions

Bone marrow & renal toxicity; cellulitis at inj site, stomatitis, alopecia, delayed erythema &/or ulceration; dyspnea w/ non-productive cough & radiographic evidence of pulmonary infiltrates; microangiopathic hemolytic anemia (hematocrit <25%), thrombocytopenia (<100,000/mm³), irreversible renal failure (serum creatinine 1.6 mg/dL); fever, anorexia, nausea, vomiting; headache, blurred vision, confusion, drowsiness, syncope, fatigue, edema, thrombophlebitis, hematemesis, diarrhea, pain.
