Indications/Uses
Parkinson's disease as monotherapy or adjunct therapy.
Dosage/Direction for Use
Monotherapy or adjunct therapy 1 mg once daily. Max: 1 mg/day. In combination w/ levodopa, w/ or w/o other Parkinson disease agents Initially 0.5 mg once daily, may be increased to 1 mg once daily. Max: 1 mg/day.
Administration
May be taken with or without food: Avoid products containing high amounts of tyramine (>150 mg), such as cheese, meats (eg, sausage or salami), bean pods, beers & soybean.
Contraindications
Concomitant use w/ MAOIs (including other specific MAO-B inhibitors), meperidine, methadone, propoxyphene or tramadol, St. John's wort, dextromethorphan
Special Precautions
Closely monitor patients w/ HTN & orthostatic hypotension; melanoma. Patients w/ mild hepatic impairment. Do not use in patients w/ moderate-severe hepatic impairment. Drowsiness. Serotonin syndrome & dyskinesia may occur w/ antidepressant & levodopa, respectively.
Adverse Reactions
Headache, dyskinesia (w/ levodopa), rash, wt loss, depression, serotonin syndrome.
Drug Interactions
Increased risk of serotonin syndrome w/ MAOIs (including other specific MAO-B inhibitors), eg, meperidine, methadone, propoxyphene or tramadol & other specific MAO-B inhibitors. Increased conc w/ CYP1A2 inhibitors & peginterferon α-2b. Increased risk of side effects eg, HTN & hypertensive crisis w/ food rich in tyramine.
