Indications/Uses
Reflux oesophagitis; acid-PUD; Zollinger-Ellison syndrome; adjunctive in duodenal ulcers associated w/ H. pylori combined w/ appropriate antibiotics; prophylaxis of acid aspiration before surgery; prevention & treatment of NSAID-associated ulcers.
Dosage/Direction for Use
Relief of acid-related dyspepsia 10 mg or 20 mg daily for 2-4 wk. Treatment of GERD 20 mg once daily for 4 wk. Refractory oesophagitis 40 mg daily. Maintenance therapy: 20 mg once daily. For acid reflux: 10 mg daily. PUD 20 mg once daily or 40 mg once daily for severe cases, continue treatment for 4 wk for duodenal ulcer & 8 wk for gastric ulcer. Maintenance dose: 10-20 mg once daily. Treatment of NSAID-associated ulceration 20 mg once daily. Prophylaxis Patient w/ previous history of gastroduodenal lesions who require continued NSAID treatment 20 mg daily. Zollinger-Ellison syndrome Initially 60 mg once daily, majority of patients are effectively controlled by doses between 20-120 mg daily. Prophylaxis of acid aspiration during general anesth 40 mg in the evening before surgery & a further 40 mg 2-6 hr before the procedure.
Administration
Should be taken on an empty stomach.
Contraindications
Hypersensitivity to omeprazole & substituted benzimidazoles.
Special Precautions
Consider possibility of malignancy before giving to patients w/ gastric ulcers as these may mask symptoms & delay diagnosis. Caution in hepatic impairment.
Adverse Reactions
Headache, diarrhea & skin rashes. Pruritus, dizziness, fatigue, constipation, nausea & vomiting, flatulence, abdominal pain, arthralgia & myalgia, urticaria & dry mouth.
Drug Interactions
May prolong elimination of diazepam, phenytoin & warfarin. Can reduce absorption of ketoconazole & possibly itraconazole.
