Pioglitazone HCl
Indications/Uses
Adjunct to diet & exercise to improve glycemic control in patients w/ type 2 diabetes (NIDDM). Monotherapy or in combination w/ a sulfonylurea, metformin or insulin when diet & exercise & the single agent does not result in adequate glycemic control.
Dosage/Direction for Use
Monotherapy in patient not adequately controlled w/ diet & exercise Initially 15 or 30 mg once daily. Patient who responds inadequately to the initial dose Dose can be increased in increments up to 45 mg once daily.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Do not use in patients w/ active bladder cancer. Use w/ caution in patients w/ prior history of bladder cancer. Not to be used in patients w/ type 1 diabetes or for the treatment of diabetic ketoacidosis. Patients in combination w/ insulin or oral hypoglycemic agents may be at risk for hypoglycemia. May result in resumption of ovulation in pre-menopausal anovulatory patients w/ insulin resistance & may be at risk for pregnancy if adequate contraception is not used. May cause decreases in Hb & hematocrit. Not to be used in pregnancy & lactation.
Adverse Reactions
Headache, URTI, myalgia, sinusitis & pharyngitis.
Drug Interactions
Increased AUC w/ gemfibrozil. Decreased AUC w/ rifampicin.
