Complicated skin & skin structure infections, complicated intra-abdominal infections in patients ≥18 yr.

Dosage/Direction for Use

Initially 100 mg, followed by 50 mg IV infusion every 12 hr over 30-60 min for 5-14 days. Severe hepatic impairment (Child-Pugh C) 100 mg followed by 25 mg every 12 hr.

Special Precautions

Increased in all-cause mortality. Anaphylaxis/anaphylactoid reactions; known hypersensitivity to tetracyclines. Monotherapy in patients w/ complicated intra-abdominal infections secondary to clinically apparent intestinal perforation. Pseudomembranous colitis; acute pancreatitis; hospital-acquired pneumonia; overgrowth of nonsusceptible organisms. May be associated w/ permanent tooth discoloration. Monitor blood coagulation parameters prior to & during treatment. May affect ability to drive & use machines. Severe hepatic impairment (Child-Pugh C); significant hepatic dysfunction & failure. Pregnancy & lactation. Not recommended in childn <18 yr.

Adverse Reactions

Nausea, vomiting, diarrhoea. Prolonged aPTT & prothrombin time, thrombocytopenia; hypoproteinaemia, hypoglycaemia, decreased appetite; dizziness, headache; phlebitis; pneumonia; abdominal pain, dyspepsia; increased AST & ALT, hyperbilirubinaemia; pruritus, rash; impaired healing, inj site reaction; increased serum amylase & BUN.

Drug Interactions

Monitor prothrombin time w/ warfarin. Affected pharmacokinetics w/ P-gp inhibitors (eg, ketoconazole, cyclosporine) & inducers (eg, rifampicin). Less effectiveness of OCs. Increased serum trough conc of calcineurin inhibitors eg, tacrolimus or cyclosporine.
