Indications/Uses
OD tab: Schizophrenia in adults; bipolar I disorders, acute treatment of manic & mixed episodes in adults. Adjunctive therapy of major depression disorders (MDD) in adults. Oral soln: Adjunct therapy for treatment of any form of parkinsonisms. Control extrapyramidal disorders caused by CNS drugs.
Dosage/Direction for Use
OD tab Schizophrenia Adult Initially 10-15 mg daily, recommended dose: 10-15 mg daily. Max: 30 mg daily. Adolescent Initially 2 mg daily, recommended dose: 10 mg daily. Max: 30 mg daily. Bipolar mania Adult Monotherapy Initially 15 mg, recommended dose: 15 mg daily. Max: 30 mg daily. Adjunct therapy to lithium or valproate 10-15 mg daily, recommended dose: 15 mg daily. Max: 30 mg daily. Childn Monotherapy or adjunct therapy to lithium or valproate Initially 2 mg daily, recommended dose: 10 mg daily. Max: 30 mg daily. MDD Adult Adjunct therapy to antidepressants Initially 2-5 mg daily, recommended dose: 5-10 mg daily. Max: 15 mg daily. Irritability related to autism disorders Childn Initially 2 mg daily, recommended dose: 5-10 mg daily. Max: 15 mg daily. Tourette's disorders Patient <50 kg Initially 2 mg daily, recommended dose: 5 mg daily. Max: 10 mg daily, ≥50 kg Initially 2 mg daily, recommended dose: 10 mg daily. Max: 20 mg daily. Oral soln Idiopathic & post-encephalitic parkinsonism Initially 1 mg daily on day 1, then increase to 2 mg 2-3 times daily for 3-5 days. Post-encephalitis 12-15 mg daily. Drug-induced parkinsonism (extrapyramidal effects) Adult Initially 1 mg, may be increased until symptoms subsides. Total daily dose: 5-15 mg.
Special Precautions
OD tab: Hypersensitivity. History of CV disease, diabetes, seizure, suicide thought. Oral soln: Discontinue use if tardive dyskinesia occur in long-term treatment. CHD, HTN, male patients w/ potency of prostate hypertrophy. Perform gonioscopic evaluation & IOP monitoring. Liver & kidney disease.
Adverse Reactions
Dizziness, nausea, constipation, headache. OD tab: Vomiting, insomnia, NMS, tardive dyskinesia, potency of metabolism changes. Oral soln: Dry mouth, blurred vision, anxiety, urinary retention, tachycardia, dilated pupils, increased IOP.
Drug Interactions
OD tab: Concomitant use w/ CNS depressants, drugs interfering w/ CYP3A4 or CYP2D6 enzymes, antihypertensive drugs. Oral soln: Antagonized w/ chlorhydria (glutamic acid, betazole). Potentiated adrenergic agents.
