Indications/Uses
60 mg cap: Wt loss in overwt adults (BMI ≥28 kg/m2) taken in conjunction w/ a mildly hypocaloric, lower fat diet. 120 mg cap: Treatment of obese patients w/ a BMI ≥30 kg/m2, or overwt patients (BMI ≥28 kg/m2) used in conjunction w/ a mildly hypocaloric diet.
Dosage/Direction for Use
60 mg cap Adult 60 mg tds. Max: 3 cap/day. 120 mg cap Adult 120 mg taken w/ water tds.
Administration
Should be taken with food: Take immediately before or during or up to 1 hr after each main meal. If a meal is missed or contains no fat, the dose may be omitted.
Contraindications
120 mg cap: Hypersensitivity. Chronic malabsorption syndrome. Cholestasis. Pregnancy & lactation.
Special Precautions
60 mg cap: Do not take >3 cap/day. May cause severe liver injury. Kidney disease. Do not take if pregnant. 120 mg cap: Closely monitor patient under antidiabetic treatment when taking orlistat. Co-administration w/ ciclosporin. GI reactions may increase w/ high fat diet. Rectal bleeding. Additional contraceptive method is recommended to prevent possible failure of oral contraception that could occur in case of severe diarrhoea. Monitor coagulation parameters while on anticoagulant therapy. Monitor renal function on patients at risk for renal impairment. History of hyperoxaluria or Ca oxalate nephrolithiasis. Epilepsy. Childn & elderly.
Adverse Reactions
120 mg cap: Headache, upper resp tract infection; abdominal pain/discomfort, oily spotting from rectum, flatus w/ discharge, faecal urgency, fatty/oily or liqd stool, flatulence, oily evacuation, increased defecation; hypoglycemia, influenza.
Drug Interactions
120 mg cap: Decrease in ciclosporin & amiodarone plasma levels. Acarbose. Oral anticoagulants. Impaired absorption of fat-soluble vit. Convulsions w/ antiepileptic drugs. Decrease absorption of iodine salts &/or levothyroxine.
