Allopurinol
Indications/Uses
Enzyme disorders, neoplastic disease, myeloproliferative disease; long-term, preventative treatment of gout; other conditions associated w/ excess uric acid in the body including kidney stones & other types of kidney disease, some tumours, certain enzyme disorders eg, Lesch-Nyhan syndrome.
Dosage/Direction for Use
Adult & elderly Initially 100-300 mg daily, may be increased depending on severity of condition. Leukaemia & certain enzyme disorders (eg, Lesch-Nyhan syndrome) Childn <15 yr 10-20 mg/kg daily or 100-400 mg daily. If dose exceeds 300 mg daily, give in divided doses.
Administration
Should be taken with food: Take after meals.
Contraindications
Hypersensitivity. Acute gout attack. Pregnancy & lactation.
Special Precautions
Gout attacks may initially increase until uric acid level stabilises. Patients w/ long-term kidney or liver problems. Concomitant use w/ mercaptopurine; ciclosporin or azathioprine; any anticoagulants eg, warfarin; chlorpropamide; vidarabine; probenecid or large doses of aspirin-type drugs (salicylates); phenytoin; theophylline; any medication for heart disease or high BP; amoxicillin or ampicillin; cyclophosphamide or other anti-cancer drugs. May affect ability to drive or operate machinery. 300 mg tab: Intolerance to some sugars.
Adverse Reactions
Skin rashes. Fever, malaise, headache, vertigo, nausea, vomiting, anaemia, uraemia, swollen glands, loss of coordination, weakness or unsteadiness, unconsciousness & unresponsiveness, sleepiness, depression, numbness, paresthesia, taste disorders or inflammation in the mouth, change in bowel habits or unusual bowel movements, impotence, sexual or infertility problems, liver problems (abnormal LFTs, hepatitis), visual problems (including cataracts), wet dream, diabetes, boils, hair loss or discolouration, heart problems & high BP, blood disorders which may be characterised by abnormal bruising, high levels of lipids in the blood, blood in the urine, swelling, breast enlargement in men.
Drug Interactions
Prolonged activity of 6-mercaptopurine or azathioprine.
