Neomycin + Polymyxin B + Gramicidin

Hypersensitivity to neomycin, polymyxin B or gramicidin.

Pregnancy and lactation.

Significant: Sensitisation (e.g. failure to heal, conjunctival and eyelid itching, reddening, and oedema), ocular effects (e.g. glaucoma, defects in visual acuity, posterior subcapsular cataract formation, secondary ocular infections) particularly during prolonged use, overgrowth of nonsusceptible organisms including fungi (long-term use).
Eye disorders: Local irritation.
Immune system disorders: Hypersensitivity reactions. Rarely, anaphylaxis.

Ophth: C

Remove contact lenses prior to administration and wait for at least 15 minutes before reinserting the lenses.

Assess for signs and symptoms of sensitisation periodically (particularly in long-term use).

Description:
Mechanism of Action: Neomycin, an aminoglycoside antibiotic, binds with the ribosomal RNA of susceptible bacteria thus inhibiting the protein synthesis and resulting in the misreading of the bacterial genetic code.
Polymyxin B is a polymyxin antibiotic. It binds to cytoplasmic membrane phospholipids of the susceptible bacteria, which alters the membrane permeability, thereby causing a leakage of essential metabolites.
Gramicidin is bactericidal against a range of Gram-positive bacteria. It binds to bacterial cell membrane forming channels through the normal lipid bilayer, thereby increasing its permeability to inorganic cations.

Store between 15-25°C. Protect from light.
