HTN either alone or in combination w/ other antihypertensive agents.

Dosage/Direction for Use

Initially 2 mg once daily taken at the same time each day, preferably in morning; may be increased to 4 mg & if necessary to 6 mg.

Overdosage

View Lacipil overdosage for action to be taken in the event of an overdose.

Administration

May be taken with or without food.

Contraindications

Hypersensitivity. Severe aortic stenosis.

Special Precautions

Patients w/ pre-existing abnormalities in SA & AV node activity; congenital or documented acquired QT prolongation; poor cardiac reserve; unstable angina pectoris; recent MI. Concomitant use w/ medications that prolong the QT interval eg, class I & III antiarrhythmics, TCAs, some antipsychotics, antibiotics (eg, erythromycin) & some antihistamines (eg, terfenadine). Patients w/ hepatic impairment. Pregnancy & lactation. No experience of use in childn.

Adverse Reactions

Headache, dizziness; palpitation, tachycardia; flushing; stomach discomfort, nausea; skin rash (erythema, itching); polyuria; asthenia, oedema; reversible increase in alkaline phosphatase.

Drug Interactions

Additive hypotensive effect w/ antihypertensive agents (eg, diuretics, β-blockers, ACE inhibitors). Increased plasma levels w/ cimetidine. Altered bioavailability w/ grapefruit juice. Reports of reversal in decreased renal plasma flow & GFR induced by cyclosporin. Possible effect on metabolism & elimination w/ CYP3A4 inhibitors & inducers. Potential interaction w/ drugs that are also protein bound to albumin & α-1-glycoprotein.
