Prednisolone
Indications/Uses
Rheumatic fever, RA, allergic diseases, nephritic syndrome, bronchial asthma & other corticosteroid-indicated conditions.
Dosage/Direction for Use
Tab Adult & adolescent 5-60 mg daily in divided doses. Max dose: 250 mg daily. Acute exacerbations of multiple sclerosis 200 mg daily for 1 wk followed by 80 mg every other day for 1 mth. Childn Adrenocortical insufficiency 140 mg/kg daily or 4 mg/m2 daily in 3 divided doses. Other indications 500 mcg to 2 mg/kg daily or 15-60 mg/m2 daily in 3-4 divided doses. Syr Adult 5-60 mL daily in divided doses or as single dose on alternate days.
Administration
Should be taken with food: Take immediately after meals.
Contraindications
Osteoporosis, active peptic ulcer, psychoses/severe psychoneurosis. Systemic fungal infections. Acute infections unless effective. Immunizations. Adjunct treatment w/ TB drugs. Pregnancy & lactation.
Special Precautions
CHF, DM, HTN, recent intestinal anastomoses, infectious diseases, uraemia, diverticulitis, myasthenia gravis, ocular herpes simplex, acute glomerulonephritis, cerebral malaria, AIDS, glaucoma, ulcerative colitis, esophagitis, epilepsy, systemic sclerosis, pheochromocytoma crisis. History of peptic ulcer or psychotic tendencies. Monitor spinal radiographs for signs of osteoporosis. RA patients & postmenopausal women. Observe for possible delayed wound healing. W/drawal syndrome. Prolonged therapy. Concomitant use w/ live virus vaccines. Hepatic cirrhosis & hypothyroidism. Chronic renal failure. Pregnancy & lactation. Growth inhibition or arrest in childn. Elderly.
Adverse Reactions
Fluid & electrolyte disturbances; vertigo, convulsions, headache, psychic disturbances; HTN, thrombophlebitis, thromboembolism, tachycardia, CHF; glaucoma, posterior subcapsular cataract; peptic ulcer, ulcerative esophagitis, diarrhea, nausea, abdominal distension, increased appetite; osteoporosis, aseptic osteonecrosis, growth disturbances in childn, myopathy; increased sweating, poor wound healing; Cushing's syndrome, acne, hirsutism, susceptibility of infections, hyperglycemia, hypokalemia, cutaneous or SC tissue atrophy, hypopigmentation, dizziness or lightheadedness, face & cheeks flushing.
Drug Interactions
Decreased effects by barbiturates, phenylbutazone, phenytoin, rifampicin, ephedrine, carbamazepine. Increased risk of GI distress, bleeding & ulcers w/ aspirin, indomethacin, NSAIDs. Increased risk of hypokalemia w/ thiazides, diuretics. Antagonized effects of hypoglycemic agents, antihypertensives. Increased risk of digitalis toxicity w/ cardiac glycosides. Altered response to coumarin.
