Donepezil HCl
Indications/Uses
Symptomatic treatment of mild to moderately severe Alzheimer's dementia.
Dosage/Direction for Use
Adult & elderly 5 mg once daily, in the evening. May be increased to 10 mg once daily after 1 mth depending on clinical response. Max: 10 mg once daily.
Administration
May be taken with or without food: Take at bedtime.
Contraindications
Hypersensitivity to donepezil HCl, piperidine derivatives.
Special Precautions
Severe Alzheimer's dementia, age-related cognitive decline. Exaggerated succinylcholine-type muscle relaxation during anaesth. Bradycardia, sick-sinus syndrome, SA or AV block. Increased risk for developing ulcers. Bladder outflow obstruction. Generalised convulsions. Exacerbation or induction of extrapyramidal effects. History of asthma or obstructive pulmonary disease. May affect ability to drive & operate machinery. Galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption. Renal & hepatic impairment. Pregnancy & lactation.
Adverse Reactions
Diarrhoea, nausea, headache.
Drug Interactions
Increased conc w/ ketoconazole, quinidine, itraconazole, erythromycin, fluoxetine. Reduced conc w/ rifampicin, phenytoin, carbamazepine, alcohol. Synergistic activity w/ succinylcholine, neuromuscular blockers, cholinergic agonists or β blocking agents.
