Dapoxetine HCl
Indications/Uses
Premature ejaculation in adult men 18-64 yr.
Dosage/Direction for Use
30 mg, may be increased to 60 mg, taken 1-3 hr before anticipated sexual activity. Consider other suitable commercial oral prep for recommended doses other than 60 mg. Do not take more than once every 24 hr.
Administration
May be taken with or without food: Swallow whole w/ water.
Contraindications
Hypersensitivity. Concomitant use w/ MAOIs; thioridazine; other antidepressants; lithium; linezolid; tryptophan; St. John's wort; tramadol; antimigraine medicines; antifungals eg, ketoconazole & itraconazole; anti-HIV medicines eg, ritonavir, saquinavir, nelfinavir & atazanavir; antibiotics eg, telithromycin; nefazodone. Patients w/ heart problems eg, heart failure or heart rhythm problems; history of fainting; history of mania or severe depression; moderate or severe liver problems. Women. Childn or adolescents <18 yr. Elderly >65 yr.
Special Precautions
Caution in patients who have not been diagnosed w/ premature ejaculation; who have another sexual problem eg, erectile dysfunction; who have history of dizziness from low BP; who use recreational drugs eg, ecstasy, LSD, narcotics or benzodiazepines; who drink alcohol; who have history of mental health problem eg, depression, mania, bipolar disorder or schizophrenia; who have epilepsy; who have history of bleeding or blood clotting problems; who have kidney problems; who have, or are at risk of, glaucoma. Perform test before treatment initiation to make sure that BP doesn't drop too much when standing up from lying down. Discuss treatment after the 1st 4 wk or after 6 doses to determine whether to continue treatment, then at least every 6 mth if treatment is continued. Wait 14 days before starting Adaporin after discontinuation of MAOIs; thioridazine; other antidepressants; lithium; linezolid; tryptophan; St. John's wort; tramadol; antimigraine medicines. Do not drink grapefruit juice w/in 24 hr prior to taking Adaporin. Avoid concomitant use w/ alcohol. Contains lactose; caution in patients w/ intolerance to some sugars. Patients who feel sleepy, dizzy, faint, have difficulty concentrating & blurred vision while taking Adaporin should avoid driving or operating hazardous machinery.
Adverse Reactions
Feeling dizzy; headache; feeling sick. Feeling irritable, anxious, agitated or restless; feeling numb or having pins & needles; difficulty getting or keeping an erection; sweating more than normal or flushing; diarrhoea, constipation or having wind; stomach pain, bloating or being sick; problems sleeping or strange dreams; feeling tired or sleepy, yawning; blocked nose; rise in BP; difficulty concentrating; shaking or trembling; lower interest in sex; ringing in ears; blurred vision; indigestion; dry mouth.
Drug Interactions
Potential interaction w/ medicines for mental health problems other than depression; NSAIDs eg, ibuprofen or ASA; blood thinners eg, warfarin; medicines for erectile dysfunction eg, sildenafil, tadalafil or vardenafil; medicines for high BP & chest pain eg, verapamil & diltiazem, or enlarged prostate; antifungals eg, fluconazole; anti-HIV medicines eg, amprenavir & fosamprenavir; antibiotics eg, erythromycin & clarithromycin; aprepitant. Increased level w/ grapefruit juice. Increased effects of alcohol.
